Force Baseline - Fitting of the calibration line
@ext.vistunou recently created the "force baseline" feature with wich the calibration line can be forced to x=0, y=0.
This, however, only affects the calibration line in the calibration plot, but not the calculation of the slope of the calibration line.
After consulting @arybina we agreed that the determination of absolute protein numbers would be more accurate if the slope of the calibration line would be calculated from the fitting in which the (0,0) intersection is forced. This influences the fit usually slightly negatively, but overall makes more sense since a low fluorescence intensity (as commonly measured in endogenously tagged cells) then does result in accurately calculated protein concentrations that are not influenced by an arbitrary y-intercept that results from fitting.
In a meeting with @ext.vistunou and @arybina, we agreed that the calculation of POIs in a segmented volume of interest can be based on the formula C = slope * (FI - mean_background) in which slope and mean_background can be extracted from the info.csv file.
